G'day Ian,
It would, the valves are two-way, which means when on deflate mode it can re-inflate. For best results, we recommend pulling the mattress out and deflating as much as possible before putting it back in your swag for pack-up. It just means you'll be able to roll your swag up a lot tighter. It's a more efficient deflation than trying to deflate and roll the swag up at the same time.
